are you guys saying SNP`s come under the heading Mutation????????,,,,,,,[single nuclotide polymorphism]

what about the repair mechanisms ?

We've started collecting fem'd beans in past couple years. We don't make seeds, just grow for smoke, and the fem'd plants we've grown (starting w/ Females seeds) have been uniform and non-herming. Saves the grow, sex, clone route. Knowing they plants are female before they get planted removes a few steps, lets us count on harvest, and makes our lives easier.

We also buy and grow reg seeds, and we have some of those we'll be doing seeds runs from (Sour 60, Blue Satellite 2.2, Blockhead to name a few). Once we've got sufficient stockpile of those beans, maybe look at making our own fem'd w/ colloidal silver, or stressing.

to my knowledge SNPs are originally a result of a mutation from a single genotype in the population say it's haplotype was AATGATTACCA and after a mutation event it became AATGATTTCCA. once this has occurred it results in a polymorphism for that nucleotide and whatever trait it might express. this happens much more than we realise but is repaired before it is able to be passed on to other generations but often if they arise it's because the repair mechanisms have failed to correct it and so it's giving to the parent's offspring. anyway sorry for getting off topic. but yeah thanks all you guys who put in info about the history of femmed seeds, some interesting stuff there

any chance you could sum-up all the different types of Mutation?

single point mutation ,,,etc?

single point mutation ,,,etc?

hey englishrick yeah sure, lol this is sorta the area i sucked at those this semester in my evolutionary genetics class but i'll do by best. there's so much on this topic so i might just stick to mutations affecting structure (also, one's affecting, function, fitness, protein sequence etc) anyway sorry for the slow reply just been trying to it up a bit simpler from lecture notes and wiki.

DNA's made up of 4 nucleotides, cytosine, adenine, guanine, thymine

DNAnucleotides.gif

Small scale mutations

* point mutations - mostly a result of chemicals or malfunction of DNA replication, exchange a single nucleotide for another. they're called transitions (changes between purine molecules A to G or G to A) or transversions (changes between pyrimidine molecules C to T or T to C). it's more common that A and G will interchange, same for C and T because they're like molecules, it's less common though happens that purines mutate to become pyrimidines, eg. A to T or T to A. but this requires a bigger molecular rearrangement. so mutations don't all have an equal likelihood of occurring!

point_mutation.jpg

- image shows a point mutation in the middle base pair (AT) it’s reversed to TA (but in mRNA T becomes U until the strands join again)

* insertions - the addition of one or more nucleotides to the DNA. These are usually a result of things called transposable elements (aka transponsons). They’re affectively “jumping genes” or mobile genetic elements (whole lines of DNA) that copy themselves and insert themselves elsewhere on the genome (can move to any chromosome). Insertions are also usually caused by errors during replication of repeating elements (AT repeats – this gets a little more complex) which cause a shift in reading frame (frameshift), both of these and transponsons can dramatically change the gene product. Insertions can be reverted by excision of the transposable element.

insertion.gif

* Deletions – the removal of one or more nucleotides from the DNA. Similar to insertions, this type of mutation can alter the reading frame of the gene. Usually deletions are irreversible though the exact same deletion could theoretically be restored by an insertion, though very unlikely. A deletion isn’t the opposite of an insertion.

Large-Scale mutations

* amplifications / gene duplications – these lead to many copies of all chromosomal regions that increases the dosage of genes located within them.

* deletions – of large chromosomal regions that leads to a loss of genes within said regions

* chromosomal translocations – the interchange of genetic parts from nonhomologous (non-sister / paired chromosomes).

* chromosomal inversions (or crossovers) – the reversing of the orientation of a chromosomal segment.

Chromosomes_mutations-en.svg

back to SNPs (single nucleotide polymorphisms) etc, they're not mutations as such, but are the direct result of mutations (as is all variation). they are a variation in DNA sequence that occurs when a single nucleotide (A,T,G, or C) in an organisms genome differs between members of a species or paired chromosomes in an individual (the individual being heterozygotic (eg. Aa) for that trait). "two sequenced DNA fragments from different individuals, AAGCCTA to AAGCTTA, contain a difference in a single nucleotide. In this case we say that there are two alleles : C and T. Almost all common SNPs have only two alleles." - wikipedia

anyway hope that helps, if it's too complex and there're parts that're hard to understand just ask me to clarify, had to read over a few parts myself to refresh it all in my head. :D

yeah for sure you should be careful with it, but this guy sounds like he

A. was drinking his own formula (DON"T DO THIS) without probably testing its strength etc
B. rubbed it on his face for a prolonged time
C. had a medical condition (Ageria?) that is linked to this discolouration since the days when silver solutions were used as antibiotics.

i'm sure it's fine to use as long as you're sensible and don't do what this guy did for many many years haha unless turning blue is what you're wanting! haha

I have to say when this threads on topic and shits not being flung its simply fantastic. Thanks guys for the serious contributions. I am glad to see this common misconception about fem seeds being bad get addressed. I have held this position before that the process should not effect the DNA. Any seeds as mentioned can be bad its all about the breeding strategy not the technique.

Thanks again guys

very true my friend!

at the end of the day it's up to the quality of the breeder, that's what defines the quality of the seeds, you can get shit seeds from M/F or F/F but the sex regime/mechanism aren't responsible for poor results. the breeder is.

Doc,,people should be careful when making statements that are bogus!!

Seriously with every post you make it seems to do a disservice to this practice...

Colloidal silver simply means silver suspended in water,,,

what is silver?

The silver I recommend using is 99.89 % pure metallic ,pharmaceutical grade.

Found in a wire form usually...

here's a cut n paste..

Silver has long been valued as a precious metal, and it is used to make ornaments, jewelry, high-value tableware, utensils (hence the term silverware), and currency coins. Today, silver metal is also used in electrical contacts and conductors, in mirrors and in catalysis of chemical reactions. Its compounds are used in photographic film and dilute silver nitrate solutions and other silver compounds are used as disinfectants and microbiocides. While many medical antimicrobial uses of silver have been supplanted by antibiotics, further research into clinical potential continues.

You cannot refer to colloidal silver as a chemical....You may however refer to sts as a chemical as it is silver broken down by nitric acid...then it becomes a chemical compound...

another cut n paste:

Silver metal dissolves readily in nitric acid (HNO3) to produce silver nitrate (AgNO3), a transparent crystalline solid that is photosensitive and readily soluble in water.

So try not to scare the people that may be reading this....

I wanted CS to be available to do reversals for two reasons

1) stable and safe,,,non toxic...(not that I was too concerned with sts as a danger).

2) anonymous method to create fems,,,,I don't advocate the ordering of these chemicals from a photo formulary store as they are turning over order info to the dea.

OK with that said,,,I'm still waiting to hear some facts from you about your claim that fem phenos will destruct and mutate as they age....especially your magic number of 5 years.....

Yes the practice of proper P1 parents is paramount to success in any breeding application....

Here's a Test run of some recent Fems made via CS by myself....

I'd feel confident stating that these are as good as you can find considering what is available today....

CS for the win! Its easy to make and works great.

Big Props Wandering Roman, it's definitely another great tool to have in the tool box. I've also had nothing but great success so far. Glad to hear Deadwood is 6 years old and you have made F1's with her and they were badass. I have some Fem Hybrid phenos from a couple lines I'm gonna continue breeding also.

very informative post. Thanks fpr posting so much info. I Personally like the fashion way of producing seed, however I do understand the usefulness of creating fem seeds. I just have to comment on the fact, from dozwns of gardens visited, that most folk still have determine if it will be a real female or produce male parts. Just doesn't seem worth it to me, well not usually.

Thanks all
