Is the 4 loci model posed by Gw Pharma legit?

Looks like the GW model has a flaw as olivetolic acid isn't built from a phenolic precursor but directly from an open chain polyketide: See 2012 PNAS publication HERE.
Should this publication be right (which sounds currently reasonable until proven otherwise), then locus 0 and A are the same.

It is likely that the route to phenolic precursors, for example, can be blocked by more than just a single gene. So it seems to be a model based on their specific population.

Genes do not block synthetic routes or whatever ;) . Genes code in most but not all cases for proteins which do or do not posses a certain catalytic activity and substrate specificity. Obviously, there is a high statistical probability that any mutation within a gene sequence of interest does lead to a null allele. In case of cannabis, null alleles such as B0 are known but extremely rare. Additionally, zero cannabinoid hemp varieties speculated to possess a defunct 0 locus are likely 'only' B0 plants. But this is a speculation as well ;) .
Besides, some genes do not code for proteins but among others for siRNA which in turn really do block 'things'. But judging from your educational level (no offence!), this is already going beyond comprehension.
Now back to your statement: Their model is in deed based on 'their specific population' which is otherwise known as the genus Cannabis ;) . The model is based on all respective knowledge on cannabinoid synthesis by the whole genus Cannabis... though only until the year 2009. Hence, they should update it...
