Fastest method to find out sex of clones

title say it all, i dont really need to root my clones, i just need to find out what sex they are, the fastest way :D

Don't know why anyone would clone a male. Are these clones from your plants? One could take clones from a parent and flower the parent to determine sex of clone still in veg.

this is outdoors and i only have one light available(2x36w neon) i didnt cut them already... but i want to eliminate or transplant males

Take a black garbage bag...select a branch of your plant and begin keeping it covered / in dark for 12 hours a day...

Might work...

have you done this? would it mess with the chemistry of the whole plant? how about just cloning that branch and putting it straight to 12/12....

yepp, the garbage bag trick works, you could also, cut off a branch, and put it in a glass of water, and give it 12 hour of light or even less... to my knowledge the fastest way. only thing else that might work is vernalisation, which means, expose the branch/clone to temperature of -5 °C for a couple of hours (or slight minus degrees), could help to induce regenerative growth

PCR (polymerase chain reaction) for the MADC sequence can identify males as early as the DNA can be harvested from leaves.

May not be practical for most, but is certainly the fastest.

Abstract

Male-associated DNA sequences were analyzed in a dioecious plant, Cannabis sativa L. (family: Moraceae), which is known to have sex chromosomes. DNA was isolated from male and female plants and subjected to random amplification of polymorphic DNA. Two out of 15 primers yielded fragments of 500 and 730 bp which were detected in all male plants but not in any of the female plants tested. These two DNA fragments were cloned and used as probes in gel blot analysis of genomic DNA. When the male and female DNAs were allowed to hybridize with the 500-bp probe, no differences in patterns were observed between male and female plants. By contrast, when these DNAs were allowed to hybridize with the 730-bp probe, much more intense bands specific to male plants were detected, in addition to less intense bands that were common to both sexes. The 730-bp DNA fragment was named MADCl (male-associated DNA sequence in Cannabis sativa). The sequence of MADCl did not include a long open reading frame and it exhibited no significant similarity to previously reported sequences.

Truly fascinating. I have no idea where to even be a part of understanding the plant on such a depth. I guess they did do the genome and it is only time before more and more such specific truths are found...

But my mind still is stuck in the dark ages I guess and I fail to even conceptualize that such research is going on...

ie.... ya just gotta stick a bag on 'er ...

dont think this will work for a hobby botanist... even if a company would offer such a "service" for hobby gardeners, ... what would be the price for a "single" evaluation??? much less "legal aspects"...

would be sweet to have that ability, but patience serves also...just sayin'.

i'll stick to both, brown paper bag and black trash bag... for my personal research of what method is better :)

so if i put already sexed females and one male of each type under a 2x36w cool white in a 14/8(maybe i can get it 20/4, but i need to transplant all plants outside before i can do that, huh) will they root? or are there small chances? i wondering if there will be any hermies out of bags? probably depends on type right? like ak47 will hermie faster than bushman in same conditions <-thats just exapaple for u to know what i mean :)

neverming guys... i read wrong... tought you said to put bags for 24/7, thats why i tought on hermies =) i just cuted clones like half hour ago, hehe. i still want to know if they will root after they show sex?:rasta:

they should root, since the plant is still alive, ... also it seems that when you cut a clone of a already flowering branch (already shows generative organs) the rooting procedure is occuring faster...
