Air Handler for one room ATTN:HVAc folks

I have a questions for you HVAC folks

So, I have a full five ton A/C unit with the compressor outside a 3000 sqft building. Inside the building the Air Handler is mounted near the ceiling about 15ft in the air.

It's not working because I'm guessing it's not charged, but that's a different story.

I want to be able to use this unit to cool a single room in the 3000sqft structure. The room is 400-500sqft, but that really doesn't matter.

I have ducting setup to cool the whole structure, but I only want to cool that room.

Is it possible to do this while running Co2? How would the air exchange work? Clearly if you're pumping air into the room, there has to be some kind of exchange.

I'm sure I'm missing something simple here. I use window units to cool the room now. I'm even considering minisplits because I want to get rid of air cooling.
 
With out the air handler being directly in the the room you will go through co2 faster than if it was in the room. You just duct the return from the room to handler.
